Because of these unique properties, dendritic materials have been intensively studied for therapeutic use.12–15 Over the past three decades since the first prototype developed in the 1970s,16 VivaGel®, the first medical product incorporating dendrimer-based therapeutic agents, has been launched, and more dendrimer-based systems are currently in clinical trials.17 In this review, we will briefly describe the structure-related properties of dendrimers and their unique synthetic strategies. The most commonly used dendrimer structures and designs will also be covered as a background to recent advances. We will then highlight the important developmental milestones and the recent formulation advances achieved in dendrimer-based nanocarriers. To conclude, we will outline the challenges that dendrimers have faced in their clinical translation and discuss how those challenges could be potentially addressed.
