It was shown previously that vancomycin production,
but not growth, can be affected by the phosphate concentration
of the medi~m.~?~~ In cultures where the
amino acids concentration was doubled at the start of
the incubation, there was a earlier depletion of phosphate
(Fig. 2A, B). If the concentration of phosphate
was increased from 2 to 5 mMsuch that the cultures were
no longer phosphate depleted, vancomycin production
could no longer be detected, confirming the negative
effect of phosphate seen in Plackett-Burman experiments.
The additional phosphate resulted in a 5% increase
in biomass levels (data not shown).
