Initially the Plackett-Burman design was used toidentify and optimize components of the semidefinedmedium that were important in stimulating specific vancomycinproduction. Further experiments examined theeffects of amino acids (selected as potential precursorsof vancomycin) on specific production. Statistical analysisof the data from the first experiment revealed thatpeptone, MgS04 . 7H20, and trace metals solution hada positive (stimulatory) effect; phosphate (as KH2P04)was identified as having a strongly negative (inhibitory)effect on specific vancomycin production (Table 11, columnA). In addition, the “dummy” variable (E) fromthe matrix were also identified as significant (Table IIA).This variable was included in the calculation of the varianceas a measure of the experimental error
