Principle of flow cytometry to sort cells. The sample of cell suspensionis introduced in the center of a narrow, rapidly flowing stream of sheath fluid. Light of asingle wavelength is focused onto this stream of cells. Each fluorescent antibody-labeledcell passing through the laser beam may get excited and emit light at a longer wavelengthwhich is detected by a number of detectors. A vibrating mechanism causes the stream ofcells to break into individual droplets. Based on the fluorescence signal, an electric chargeis placed onto the drop containing a cell which then gets deflected and sorted into differentcollection tubes by an electrostatic deflection system.
