Hairy roots were obtained after 19 weeks past transformation
of Taxus  media var. Hicksii with the
agropine-type of Agrobacterium rhizogenes strain
LBA 9402. The other tested strains, i.e., ATCC
15834 and TR 105 did not induce the formation of
hairy roots. The transformation frequency was 3%
of seedlings. The 2-cm long roots were excised and
cultured individually in liquid DCR medium. During
the first year of cultivation the growth of the hairy
roots was poor. Finally, after 1.5 year, the stable culture
was achieved. However, callus formation was
observed, occasionally. Hairy roots grew well in modified
DCR liquid medium without growth regulators,
every 4 weeks 0.3 g of inoculum was transferred to the
30 ml of fresh medium. The pH value of the medium
was 3.8 and the level of sucrose was 3%. The conductivity
of the medium at the beginning of culture
was 4 m s..1 and it started to decline since day 20 of
culture while the pH started to increase. The content of
sucrose started to decline at the same time as conductivity.
The hairy roots were long, well branching and
formed root hairs typical of transformed root culture
