Precipitation of Protein by ammonium sulfate
The crude fungal culture was filtered through Whatmann No 1 filter paper using a vacuum pump and the resulting
filtrate was concentrated kept at 4o C. Proteins were precipitated by ammonium sulfate from the mycelia filtrate
obtained after submerged culture fermentation. Solid ammonium sulfate was added slowly to the crude extract of each
isolate to give 80% saturation and the solution was stirred gently for at least 1 hour at 40
C and then left to stand over
night. The precipitate was collected by centrifugation at 8,000 g for 1 h at 40
C. The supernatants were discarded and
the pellets were dissolved in Minimum amount of 5mM Bis tris (Bis – 2 – hydroxy ethyl) imino tris (hydroxy
methyl ) methane) HCL buffer pH 6.5.
