Pederin blocked mitosis of normal and tumoral
mammalian cells in vitro at concentrations
of 1 ng/ml (Soldati et al. 1966). Age responses
of cultured mammalian cells to pederin
were similar to responses to another DNA inhibitor,
hydroxyurea, when the latter drug was
10,000 times more concentrated (Mauro & Madoc-Jones
1970). Use of radioactive precursors
to analyze macromolecular synthesis showed that
pederin blocks protein and DNA synthesis almost
immediately without affecting RNA synthesis
(Brega et al. 1968). Suggestions that pederin
may inhibit translocation (Jacobs-Lorena
et al. 1971) were supported in studies of human
tonsil ribosomes by Carrasco & Vazquez (1972).
Earlier studies prompted use of pederin in mitochondrial
protein synthesis research by Brega
& Baglioni (1971) and Brega & Vesco (1971).
In experiments on segmentation mitoses in
newt eggs, pederin blocked DNA synthesis,
causing anomalies at telophase and stopping nuclear
evolution between telo-prophase and true
prophase (Sentein 1973, 1975; Sentein & Ates
