The concentration of total cholesterol in serum has been associated withmetabolic, infectious and coronary heart diseases. In the plasmacholesterol is transported by three lipoproteins: high density lipoprotein (HDLCholesterol), low density lipoprotein (LDLCholesterol), andvery low density lipoprotein (VLDL- cholesterol).Castelli and co-workers have indicated that an inverse relationshipexists between serum HDl-Cholesterol and the risk ofcoronary heattdisease. The measurement of total and HDL Cholesterol andtriglyceride provides valuable information for the prediction ofcoronary heart disease and for iipoprotein phenotyping.Serum is reacted with a detergent which is selectively absorbed bynon-HDL iipoproteins, ie., LDL, VLDL and Chylomicrons. Thus, onlyflDL lipoproteins are available to be solubilized and released to reactwith the cholesterol esterase, cholesterol oxidase and chromogens (togive color).
