Azan-stained sections of the kidney fixed in 4 different fixatives: F, 10% neutral buffered formalin; D, Davidson’s solution; B, Bouin’s fluid; H, Helly’s fluid. All panels are shown at the same magnification. These tissues were taken from the same individual rainbow trout, Oncorhynchus mykiss. It is rather well known that simple formalin fixation is not suitable for Azan staining, which is also apparent in this figure. Although the best result seems to have been obtained with the tissue fixed in Davidson’s solution at first sight, both the preservation of fine tissue morphology and differential staining of tissue components are actually better in the tissue fixed in Helly’s fluid. All sections were cut at 2 μm.
