Culture conditions
Spores of A. orientalis were inoculated into a 500-ml.Erlenmeyer flask containing 50 ml growth medium and shaken at 30°C for 48 h. One milliliter of the culture was added to a 500-ml baffled flask containing 100 ml seed medium and cultivated at 30°C and 250 rpm for 60 h. This seed culture was then transferred into a baffled flask orfermenter. Flask experiments were performed in a 500-ml baffled flask containing 50 ml production medium, at 34°Cand 250 rpm for 120 h. The initial pH of the production medium was adjusted to 7.0. Fermenter experiments were performed in a 7-l jar fermenter (Kobiotech, Incheon,Korea) containing 4 l production medium. Temperatureand pH were controlled at 34°C and 7.0, respectively;agitation speed was maintained in the range of 500–900 rpm, to keep the dissolved oxygen concentration above20%. DOT was monitored using Ingold polarographic DO electrode (Mettler Toledo). The pH values of culture brothswere monitored using a pH-meter (Mettler Toledo) and controlled by ammonium hydroxide (30%). The aeration
rate was 1.0 vvm during fermentation.
