What is the optimum temperature for the peroxidase reaction?
Many enzymes have a temperature optimum that is similar to the optimum growth temperature
for the organism. Does peroxidase? If it does, it may be a result of adaptation by natural
selection. But, what if it does not? This experi
ment will give you the data from which to argue
for, or against, the idea that peroxidase has undergone direct selection for the optimum reaction
temperature.
This experimental approach would be simplif
ied if you and the spectrophotometer could be
placed in a room with the appropriate temperat
ure; however, ice-bath temperatures and 55°C
would both be uncomfortable—to say the least! So, what you will do is to perform the reaction
at the temperature of interest for exactly two minutes, then, very quickly, move the tube to the
spectrophotometer to determine the absorbance at 655nm for a single 2-minute time point.
