Tissue-engineered skin has been in clinical use for 25 years andhas developed greatly during this time. This review looks at the rolebiomaterials need to play in providing for epidermal cover, dermalreplacement, and epidermal/dermal replacement, and describes themajor problems that remain. The majority of biomaterials in clinical useare based on natural or extracted collagen. The clinical challenges inusing these materials are highlighted throughout – specifically safetyissues, improving the take of cultured cells on wound beds, improvingthe rate of neovascularization of tissue-engineered skin, and developingscaffolds that resist contraction and fibrosis.
