Healthy apical shoot buds or young shoot cuttings of V. negundo L. each bearing four to five nodes were collected during various months from a mature plant growing in the Aromatic and Medicinal Plant
Garden of the Regional Research Laboratory at Bhubaneswar. After removing the leaves, the shoot apices and nodal segments (0.6–0.8 cm), the latter with dormant axillary buds, were washed thoroughly under running tap water for 45 min followed by treatment with a solution of 5% (vol/vol) Laboline (Qualigens, India) and 7% (vol/vol) sodium hypochlorite (BDH, India) for 7–10 min and
thereafter washed five to seven times with autoclaved water. The explants were then surface-disinfected with 0.1% (wt/vol) aqueous mercuric chloride solution for 8 min and finally rinsed with autoclaved
double-distilled water (six or seven changes).
