Materials. Commercially available fenugreek seeds of different geographical origins
and fenugreek oleoresin were used. Sotolone was from Aldrich and diethyl 2-methyl-
3-0xobutanedioate~ L-isoleucine, methylglyoxal (40% in water), phenylglyoxal, 2,3-
butanedione, and 2,3-pentanedione from Fluka. Solvents and other chemicals were of
analytical grade from Merck.
Synthesis. 3-Amino-4,5-dimethyl-3,4-dihydro-2(5H)-fbranone hydrochloride (ADF)
and 4-hydroxy-L-isoleucine (HE) were obtained as diastereomeric mixtures by
photochemical chlorination of L-isoleucine (Figure 2a), i.e. (3S,4R,5RJ3S,4R,5S) and
