Mitochondrial metabolism of pyruvate may also regulate the cytosolic
NADH/NADþ ratio indirectly. The cytosolic NADH/NADþ ratio is set by the ratio of lactate to pyruvate in the cytosol and the steady‐state level of cytosolic pyruvate is balanced both by the flux of pyruvate from the medium and by the import of pyruvate into the mitochondria (Dumollard et al., 2006b). Such import of pyruvate into the mitochondria is necessary to maintain a reducing cytosolic redox potential as blocking pyruvate entry into mitochondria induces a sharp LDH‐dependent oxidation of cytosolic NADH (Dumollard et al., 2006b).
