Schematic illustration of the monitoring principle of the ampli-
fication process by RT-LAMP using MB. Reverse transcription and
DNA amplification can be accomplished at a constant temperature (63
C) in a short time in one-step by the use of reverse transcriptase together
with Bst DNA polymerase. The electroactive reagent MB binds to
amplified DNA. Hence, the peak current of reaction mixture decreases
with the amplification because of the slow diffusion of MB-amplified
DNA complex to the electrode surface. The peak current measured by
SVW decreases with the amount of amplified DNA.
